Рассчитанная методом регрессионного анализа среднеэффективная доза бемитила на данной модели составила 210 мг/кг, среднеэффективная доза мелатонина 99 мг/кг. В составе комбинаторного сочетания в соотношении 5:1 активность соединений существенно возрастает, ED50 бемитила снижается до 85 мг/кг, аналогичный показатель мелатонина становится 14,7 мг/кг. Назначение комбинации 100 мг/кг бемитила и 20 мг/кг мелатонина мышам приводит к увеличению продолжительности их жизни в стандартных условиях гипоксии до 46 ± 4,3 минут, что на 138,5 % больше аналогичного показателя животных, получавших плацебо. Анализ фармакодинамики комбинации препаратов по T. Chou показал аддитивный тип взаимодействия, что позволяет снижать дозы компонентов при их сочетанном применении в 2,0–6,7 раз. Ввиду отсутствия видовой специфичности действия бемитила и мелатонина их синергичная комбинация может быть интересна для применения у человека.</p></abstract><trans-abstract xml:lang="en"><p>The antihypoxic effect of a combination of bemethyl and melatonin after single intraperitoneal injection was studied in a model of hypercapnic hypoxia in a hermetic volume in mice. The average effective dose of bemethyl in this model, calculated by regression analysis, was 210 mg/kg, the average effective dose of melatonin was 99 mg/kg. As part of a combination in a ratio of 5:1, the activity of the compounds increases significantly, the ED50 of bemethyl decreases to 85 mg/kg, the same indicator for melatonin becomes 14.7 mg/kg. Administration of a combination of 100 mg/kg of bemethyl and 20 mg/kg of melatonin to mice leads to an increase in their lifespan under standard hypoxic conditions up to 46 ± 4.3 minutes, which is 138.5 % more than in animals receiving placebo. An analysis of the pharmacodynamics of the combination of drugs according to T.
